چکیده

Abstract Background Lipidome disturbance has long been recognized to occur after myocardial infarction (MI). Accumulation of excessive fatty acids induces production reactive oxygen species and consequently deteriorates cardiac injury in MI. However, the spatial temporal lipid profile heart following ischemic remains unknown. Purpose We aim uncover temporal-spatial lipidome ischemia reperfusion (I/R) identify circulating lipids released from injured myocardium that are potentially useful for diagnosis disease. Methods C57/BL6 mice were subjected 30 min followed by removal ligature establish injury. Porcine I/R was induced 105 reperfusion. Human plasma obtained 143 post-MI patients. Myocardial profiles generated matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MALDI-MSI) different regions (infarct, remote peri-infarct) at time points. Moreover, analysed LC-MS/MS. Results observed a drastic alteration with distinct spatial-temporal features both MALDI-MSI In infarct tissue, as revealed LC-MS/MS, we an elevation glycerolipids peaked 3 hours I/R, sustained phospholipids sphingolipids up days. Similar alternations but much weaker peri-infarct tissue compared tissue. Among those lipids, PC 32:0 detected highly overlapped CD68 staining single-cell level, showing strong correlation macrophage infiltration mouse hearts (R2=0.93, p<0.0001). A similar increase area also porcine Surprisingly, levels decreased humans, patients lower than healthy individuals (p=0.03). Further analysis demonstrated determined within 72 percutaneous coronary intervention negatively correlated 6-month ejection fraction (R2=0.08, p<0.001). Conclusions established synergizing LC-MS/MS imaging. positively function Our findings indicate is potential biomarker inflammatory status heart. Myocardial lipidomics. Developments in myocardial nuclear lipidomics.

The development of electrospray ionization mass spectrometry has been critical for the analyses of lipidomes from subcellular organelles. The myocardial nuclear lipidome likely has a key role in the molecular regulation of gene expression. In fact, recent studies have suggested that specific phospholipid classes bind and regulate specific transcription factors. The dynamic regulation of the myo...

Measuring spatial - temporal of Yazd urban form using spatial metrics

Abstract Urban form can be affected by diverse factors in different times. Socio- economic, political and physical factors are among the main contributors. So, one of the most important challenges of urban planners is measuring and identifying urban development pattern in order to direct and strengthen it to sustainable pattern and right direction. The case study of the present paper is the ...

Spatial organization of acute myocardial ischemia.

INTRODUCTION Myocardial ischemia is a pathological condition initiated by supply and demand imbalance of the blood to the heart. Previous studies suggest that ischemia originates in the subendocardium, i.e., that nontransmural ischemia is limited to the subendocardium. By contrast, we hypothesized that acute myocardial ischemia is not limited to the subendocardium and sought to document its spa...

Osteoprotegerin levels in ST-elevation myocardial infarction: Temporal profile and association with myocardial injury and left ventricular function

BACKGROUND Elevated levels of osteoprotegerin (OPG) have been associated with adverse outcomes in ST-elevation myocardial infarction (STEMI). However, the role of OPG in myocardial injury and adverse remodeling in STEMI patients remains unclear. The aims of this observational cohort study were to evaluate: 1) the temporal profile of OPG during STEMI, 2) possible associations between OPG measure...

Temporal profile of protein release in myocardial infarction

Acute myocardial infarction (AMI) is one of the most important cardiovascular diseases in developed countries, where it represents the major cause of death and disability. The diagnosis of ‘‘classic’’ AMI is based on the triad of typical symptoms and changes in the electrocardiogram and in biomarkers. Biomarkers are proteins that are released into the circulation by the damaged myocardial cells...
